生化机理：TW-37 is Bcl-2 inhibitor (Ki values are 0.29 μM and 1.11 μM for Bcl-2 and Bcl-XL respectively). TW-37 inhibits the angiogenic potential of endothelial cells in vitro. TW-37 induces S-phase arrest and apoptosis in pancreatic cancer cell lines; also inhibits the activation of Notch-1 and Jagged-1 in vitro and in vivo.Small-molecule inhibitor of BCL-2 ( K i = 0.29 μM). Inhibits multiple BCL-2 family members including BCL-XL ( K i =\xa01.11\xa0μM) and MCL-1 ( K i = 0.26 μM). Binds to the BCL-2 homology domain 3 (BH3) groove of BCL-2.

Western blot analysis showed that the expression level of Notch-1 was significantly lower in tumors from the TW-37–treated mice than those from vehicle-treated control mice. In addition, the expression of Jagged-1 and Notch-1 downstream target gene, Hes-1, was also down-regulated in TW-37–treated tumors.
